摘要利用二酯法化学合成了一个脱氧八核苷酸(d-G-G-A-A-T-T-C-C)。此八核苷酸片段自身互补形成双链DNA片段，能被限制性内切酶EcoRI酶解。利用双向同系层析测定了它的核苷酸序列，证明这一片段的序列符合实验设计要求。
